Pokey Blocks are objects that appear in Super Skywheel in Mario vs. Donkey Kong: Mini-Land Mayhem!, being introduced in Area 8-3. They also reappear briefly in Rainbow Summit, specifically in Area S-16. They are static yellow spotted squares with dotted borders that appear in groups of three, either horizontally, vertically, or with two blocks protruding out of the center block from different directions to form an "L" shape. Pokey Blocks can be placed with any of these configurations in the Construction Zone. Minis can pass through Pokey Blocks while they are not filled in; however, if a Pokey moves into the Pokey Blocks, each segment of the Pokey will be absorbed into each Pokey Block, which permanently turns the Pokey into three harmless and immobile blocks that act identically to ground. This allows the Pokey to be used as a platform or wall by the Minis, as well as a staircase if the Pokey Blocks are configured in an "L" shape.

Notably, Pokeys trapped in Pokey Blocks use the same sprites as when they turn into blocks after the use of a yellow color switch in Mario vs. Donkey Kong: Minis March Again!. A visual difference between the two games is that when Pokeys turn into horizontally oriented blocks, in Minis March Again!, the Pokey always uses its face segment as the center block, whereas in Mini-Land Mayhem!, the Pokey's face segment is always either the left or right block. The only exception to this is in Area S-16, where the Pokey trapped in the horizontally oriented Pokey Blocks will always use its face segment as the center block.
