Lava Dome is the fifth floor in Mario vs. Donkey Kong 2: March of the Minis. Volcanoes and a smoke-filled night sky can be seen in the background. The music tracks played on this floor are remixes of the Bowser's Castle theme from Super Mario Bros.; additionally, this floor bears a significant resemblance to worlds concerning Bowser found in various other Mario games.

This floor features fire-based obstacles such as lava, Lava Fountains, Podoboos, and Fire Bars. Spy Guys are introduced on this floor, while Circus Kongs, Crash Kongs, and Cool Kongs return from previous floors. This floor also introduces Wall Jump Bars, Fire Blocks, and wood platforms, which can be burnt to access new areas.

The battle with Donkey Kong in Room 5-DK, referred to as Boss Game 5, plays similarly to Boss Game 1 in Room 1-DK. However, instead of dropping barrels and dumbbells, Donkey Kong will slam the ground to drop fireballs from above. After taking a hit, Donkey Kong will drop a large amount of fireballs, as well as a water droplet. If a Mini Mario comes in contact with a water droplet, it gains a shield that protects it from fireballs and Fire Bars while in the air. However, if the Mini is hit by a fireball while still in the cannon, the shield disappears. If all of the player's Mini Marios are destroyed, the game is over. After taking 6 hits, Donkey Kong is defeated and flees to Toadstool Castle.

Upon completing Room 5-DK, the Lava Dome construction kit is unlocked for the Construction Zone.
