Toadstool Castle appears as the sixth floor of Mario vs. Donkey Kong 2: March of the Minis. This floor introduces a variant of elevators known as Moving Platforms, as well as Balance Beams. Both of these objects can be stood on by Mini Marios to reach otherwise inaccessible areas. Nearly all of this floor's levels also prominently feature Color Switches. The music tracks played on this floor are arrangements of "Inside the Castle Walls" from Super Mario 64.

No new enemies are introduced on this floor; however, Shy Guys and Thwomps return from previous floors. Additionally, every previously introduced monkey robot, including Circus Kongs, Capture Kongs, Crash Kongs, and Cool Kongs, appears on this floor at least once.

The battle against Donkey Kong in Room 6-DK, referred to as Boss Game 6, is similar to Boss Game 2 in Room 2-DK and Boss Game 4 in Room 4-DK. However, the spiked platform Donkey Kong is on both moves left and right and sways in an arc. Donkey Kong also slams the spiked platform more often than during Boss Game 4, spawning Spiked Balls from the spiked platform's location more frequently than in Boss Game 4. To defeat Donkey Kong, the player must shoot Mini Marios from a cannon to either hit Donkey Kong directly, or reach the platforms on the top of the screen and cause Toad portraits to fall on Donkey Kong. However, these portraits can also destroy a Mini Mario if it comes into contact with one. If all the player's Mini Marios are destroyed, the game is over. After Donkey Kong is damaged, his spiked platform briefly sways violently. After Donkey Kong takes 6 hits, he is defeated and flees to Spooky Attic.

Upon completing Room 6-DK, the Toadstool Castle construction kit is unlocked for the Construction Zone.
