Toadstool Castle appears as the sixth floor of Mario vs. Donkey Kong 2: March of the Minis. This floor's levels are the first in the game to feature moving platforms, and balance beams similar to the ones from Super Mario Bros.

No new enemies are introduced into this area, but all the toy Kongs appear as enemies at least once. Thwomps are plentiful as well. This area even introduces the Balance Beam of which the Minis get on to lower it. The more Minis on one end, the lower that platform goes.

The battle against Donkey Kong on this floor, referred to as Boss Game 6, is similar to Boss Game 2 in Room 2-DK and Boss Game 4 in Room 4-DK. However, the spiked platform Donkey Kong is on both moves left and right and sways in an arc. Donkey Kong also slams the spiked platform more often than during Boss Game 4, spawning Spiked Balls from the spiked platform's location more frequently. To defeat Donkey Kong, the player must shoot Mini Marios from a cannon to either hit Donkey Kong directly, or reach the platforms on the top of the screen and cause Toad portraits to fall on Donkey Kong. However, these portraits can also destroy a Mini Mario if it comes into contact with one. If all the player's Mini Marios are destroyed, the game is over. After Donkey Kong is damaged, his spiked platform briefly sways violently. After Donkey Kong takes 6 hits, he is defeated and flees to Spooky Attic.

The music that plays in the levels is an arrangement of "Inside the Castle Walls" from Super Mario 64.
