They are capable of speech, as they talk to the player when their character runs into them. Each of them often uses words that start with the same letter as its note. They offer a [[coin]] trade for a [[Star (Mario Party series)|Star]]. They seem connected to the place, yet they do move around willingly across the entire place whenever a metronome is ticking.

The Musical Notes are five sentient eighth notes that appear above Star Spaces in Toadette's Music Room in Mario Party DS. When reaching a Star Space, the present Musical Note will offer a Star in exchange for coins. Each note will require a different amount, ordered from least to most expensive; the blue, green, yellow, orange, and red notes will accept 5, 10, 15, 20, and 30 coins respectively.

When the Star Space is relocated, the next Musical Note in the cycle will appear above it. Moving the Star Space with a Mysterious Metronome allows Musical Notes to be skipped without anyone purchasing a Star.

Daisy encountering Musical Note B (NA release)

The Musical Notes' individual identities are contradictory between the North American and European versions of the game. The NA names are a reference to the lines of the treble clef staff, from bottom to top (E-G-B-D-F). Instead, the EU version has them named after the first five notes in the C major scale (C-D-E-F-G). This results in dialogue for Musical Notes B and C being exclusive to the NA and EU versions respectively. Similarly, the Japanese and Korean versions designate names based on Western solfège (Do-Re-Mi-Fa-Sol).
